Elavil and SAM-e. Same chemical makeup?

I have been going through the archives, reading till my eyes and brain ache! Can anyone tell me if the above mentioned two achieve the same thing?

Correct me if I'm wrong but Elavil targets serotonin and norepinephrine (a weak norepinephrine?), right? As stated before, this drug works for me to help me sleep, helps anxiety and depression.

What does SAM-e target? Hopefully if it targets the same as Elavil, I would like to try it. If not, I'll try something else that goes along the same lines as the Elavil; which could be doxepin, trimipramine or nortriptyline (ideas??). As stated in another post of mine, the SSRIs don't agree with me (anxiety, aggressiveness, insommnia).

Hopefully, someone more knowledgable in the field of drugs (or experience) will help me out.

James here....

Doxpin is the most sedating of all the TCA's, so you might
try the other substutions trimipramine or nortriptyline. I also
used Sumontril for a while and liked it. The other options that come to mind
in keeping with effecting nor-e and 5ht are: Remeron or Effexor. Remeron is related
to the tricyclics, Effexor is a novel compound. I would
think Remeron would be a good choice, as you like TCA's.
It has all the good traits of a TCA (robust AD effect, sedation) w/o the anticologernic and
most of the antihistaminic (side)effects.

How does SAM-e do it? SAM-e, or S-adenosyl-L-methionine, is a naturally
occurring molecule found in virtually all body tissues and fluids. It acts as a
"methyl donor" in biochemical reactions and is therefore involved in producing
and processing a wide range of chemicals that maintain normal cell function.
And because it's a naturally occurring substance, SAM-e appears less likely
to cause adverse side effects than synthesized drugs. Its most common side
effect is stomach upset.

S-adenosylmethionine is a small molecule found in all organisms. Its primary function is as a methyl donor in the synthesis of RNA/DNA, protein, polyamines, and phopholipids. Thus, SAM is in every part of the body and is a common component of cellular function. Given that, defining a specific target is very difficult. The only correlation to depression that I know of is that SAM may increase levels of many amines including serotonin, norepinepherine, and dopamine. However, there is evidence that SAM is not taken by intact cells. Personally, I would be very suspect of SAM as an antidepressant, and I would not use it instead or mix it with prescribed medications.
