Though the exact mechanism is not known, there is some evidence that desipramine hits the dopamine D2 receptor. I took amisulpride for a while, which acts on the D2 receptor, so I am familiar with what it felt like. Desipramine had that same kind of feel to it, in terms of motivation and activity. But wow, the side effects of the TCAs are just too much for me.

All of the TCAs are so dirty, hitting so many different receptor types, that it seems to me they must all affect dopamine in one way or the other. Probably not direct reuptake inhibition though.
