A phase 3 clinical trial for postpartum depression is expected to be released in January. If the trial is a success, SAGE-217 could receive FDA approval for PPD by late 2019. Sage is recruiting now for their phase 3 trial for major depressive disorder. If this trial is a success, SAGE-217 could receive FDA approval for MDD in 2020 or 2021. However, if SAGE-217 is approved for PPD in 2019, physicians would be able to prescribe it off-label for MDD, bipolar I/II disorder, and insomnia.

In the clinical trials, it has performed significantly better than placebo for anxiety.

Sage-217 is an interesting one. Many of the SSRIs indirectly target the same neurosteroid pathway. As a positive gaba-a modulator, it could also replace the use of benzodiazapines for some people.
I have been benefitting from CBD which is also a positive modulator of the gaba-a receptor.Linkadge

48 years of benzos now low dose one time a day any hope of even getting off the benzos. Wow can't believe how old I am now. And I often wonder if taking them is responsible for loss of taste and smell 15 years now. And I really don't feel the benzos work any longer. It seems if something is bothering me I just don't sleep even with them. Phillipa

Its not unusual to not feel benzos anymore after a while. I am amazed that they worked for so long. If i take like 2-3 Ativans, i dont feel it anymore. I must not take it for peanuts in order for it to work in an emergency.

This is why I am pretty sure they don't anymore. Wonder what would happen if just stopped them since two one at a time? Sometimes I cut the dose and nothing happens? Phillipa

I would be careful and taper gradually. Sometimes withdrawal symptoms are delayed. You may get sick if you just stop. Id do 10% steps with dissolved benzo.

I take 7.5mg of valium a night and then .75mg of xanax? Is this high? Phillipa

Its not much but your brain is likely used to that alot. You wont do yourself a favor to just quit abruptly. I think if your will is strong to get off you can do it. I always recommend the website https://www.survivingantidepressants.org
They have lots of experience with withdrawing. Be warned though, they dont like meds.

This is true of Abilify. Some people don't get withdrawal effects until 3 weeks after their last dose. Anxiety is a big one.

This Sage-217 is a mystery to me. What is the mode of action? Does it work for anxiety? Is it ok when you are inclined towards psychosis? What are side effects and risks?

"Its mechanism of action is unknown, but it appears to be a positive allosteric modulator of GABA-A receptors with a binding site distinct from benzodiazepines."
The above quote is from the FDA Briefing Document for brexanolone, the intravenous form of SAGE-217.
