I've been following the company Alkermes for a couple of weeks as I had great success with buprenorphine for treatment resistant depression with generalized anxiety via it's kappa "antagonism/blocking" receptor effects (my best hypothesis based on having had other morphine related medications in either the hospital or ER; mu receptor opioid agonists like Vicoden's receptor effect and Demerol's kappa opioid "agnonist" receptor effect and now the company Alkermes has Alkermes 5461 (buprenorphine plus ?) going into phase II (http://www.alkermes.com/Research-and-Development/Pipeline) for treatment resistant depression as well as for cocaine addiction.
Alkermes 5461 is buprenorphine plus either a new opioid blocker like naltrexone or it may be Low Dose Naltrexone which normalized endorphin levels and prevents addiction to the buprenorphine and Alkermes owns the long acting liquid naltrexone Vivitrol so my guess is maybe the second component of AlKS 5461 is naltrexone, but Alkermes might have another chemical in development. WOW this is VERY cool, had no idea someone was actually looking at using it for TRD in terms of clinical development. i wonder where someone could get in to a trial if available? i guess clinicaltrials.gov or have you seen it elsewhere talked about? sorry english bad tonight.

I haven't even looking for the clinicl trial on clinical trials dot gov for ALKS 5461 (buprenorphine plus ALKS 33) but I looked again at Alermes website, and it's just finished phase 1 (no press release) but can only find information for 1/2 of phase 1

"ALKS 5461" is the combination buprenorphine plus that of ALKS 33 and buprenorphine. ALKS 33 has already finished phase II and going to phase 3. ALKS 33 is going to be indicated for alcohol addiction but didn't pass for binge eating.

Not sure if any conclusions yet except that the key will be the ALKS 33 study as I already know buprenoprhine works for me (hope it works for the study) and from what I can see on this study, they exclude anyone allergic to naltrexone making me think it's low dose naltrexone which is already used by psychiatrist given at night to increase natural levels of endorphins because the receptor overcome the small dose therefore increasing endorphins. Low Dose Naltrexone is used off lable for many autoimmune diseases like Huntington's, MS, etc which good success.

The Phase 2 of ALKS5461 is now completed and successful results have already been reported.
Does anyone know when this drug is estimated to be launched?
I have had good results with Naltraxone and Subutex separately and I really think that this is going to revolutionize everything.
I have even written to the company but they have not responded.
