There's a new Anti-depressant out in the EU and Canada called Valdoxan. I need help knowing from you experts out there more about it's mechanism of action. I don't understand the agonist/antagonist thing.
Anytime I take something that affects Serotonin, I respond with increased anxiety, which is a problem because I'm trying to treat GAD. I've tried all the SSRI's. Since this has a unique mechanism of action, I'm having a hard time figuring out how this Valdoxan works and how I would respond.
Valdoxan.com says that Valdoxan is a melatonergic agonist (MT1 and MT2 receptors) and 5-HT2C antagonist. Valdoxan increases noradrenaline and dopamine release specifically in the frontal cortex and has no influence on the extracellular levels of serotonin.
It says that it's a 5HT2c antagonist but it doesn't affect extracellular serotonin. That seems like a contradiction. If I don't respond well to things that affect serotonin, will I react badly to this?
I'm assuming here, of course, that I can buy this drug from Canada.

I didn't know it was in Canada. I also don't understand agonist vs antagonist although as been explained to me. The sites I read say helps withs circadiam rhythms so helps with sleep and no sexual effects. But then compared it to paxil and zoloft. I also get anxiety with any ad. Great question. Phillipa

Hi Phillipa,
I think that Agonists Stimulate receptors and antagonists block them.
